摘要: P53基因是迄今发现与人类肿瘤的发生相关性最高的抑癌基因,在人类癌症中发生突变的频率高达50%。在子宫颈癌组织中表达则降低,且以野生型(wt)为主。wtP53第4外显子72密码的基因多态性与子宫颈癌发病机制密切相关。wtP53蛋白在子宫颈癌细胞中半衰期短,功能易失活,具有调节细胞周期、促进肿瘤细胞凋亡、抑制肿瘤细胞生长的功能。其功能失活与多种基因,如E6,E7,MDM2和STAT3等基因的降解作用有关。

Abstract: P53 gene is one of the most important human tumor suppressor genes relative to tumorigenesis,and its mutations are found in approximately 50% of cancers. Though P53 is expressed lower in cervical cancer tissues with the main wild-type. The codon 72 polymorphism in exon 4 of wtP53 is closely associated with occurrence of cervical carcinomas. In cervical cancer cells the half-life of wtP53 protein is shorter, and the function is inactivated. wtP53 regulate of cell cycle, promotion of tumor cells apoptosis, and inhibition of tumor cell growth. A variety of genes such as E6, E7, MDM2, STAT3 can inactivate wtP53 and induce the degradation of wtP53.
