Expression and Clinical Significance of BRMS1 and NF-κB in Cervical Carcinogenesis

摘要: 目的:探讨乳腺癌转移抑制基因1(BRMS1)、核因子相关κB结合蛋白50( NF-κB p50 )在宫颈癌变过程中的表达及其临床意义。方法:采用免疫组化(Super Sensitive TM IHC)法分别检测慢性宫颈炎组织、CINⅠ组织、CINⅡ~Ⅲ组织和宫颈癌组织中BRMS1、NF-κB p50 的表达,分析两者的表达水平与宫颈癌临床病理特征的关系。结果:①宫颈癌、CINⅡ~Ⅲ及CINⅠ组组织中BRMS1蛋白的表达低于慢性宫颈炎组,差异有统计学意义(均P<0.05),且BRMS1蛋白在ⅡB~Ⅳ期、浸润深度>1/2、有淋巴结转移者中表达降低甚至缺失(均P<0.05)。②NF-κB p50蛋白在宫颈癌组、CINⅠ和CINⅡ~Ⅲ组中的阳性表达率高于慢性宫颈炎组(均P<0.05),且在瘤体直径≥4 cm、低分化癌、浸润深度>1/2、有淋巴结转移者中NF-κB p50蛋白表达升高,差异有统计学意义(均P<0.05)。③宫颈癌中BRMS1与NF-κB p50蛋白表达水平呈负相关(rs=0.426,P<0.001)。结论:宫颈癌中BRMS1蛋白与NF-κB蛋白的阳性表达呈负相关,BRMS1蛋白阳性表达逐渐下降、甚至缺失,而NF-κB蛋白阳性表达逐渐升高,与癌细胞的发展、浸润转移密切相关。

Abstract: Objective:To study the protein expression and clinical significance of breast cancer metastasis suppressor l(BRMS1) and nuclear factor kappa B(NF-κB p50) in cervical carcinogenesis. Methods:The immunohistochemical method(Super Sensitive TM IHC)was used to determine expressions of BRMS1 and NF-κB p50 were in chronic cervicitis tissues,CINⅠ tissues,CINⅡ-Ⅲ tissues and cervical carcinoma tissues. The clinical significance was analyzed with histopalthological records. Results:① The positive rates of BRMS1 in cervical carcinoma ,CINⅠand CINⅡ-Ⅲ tissues were much lower than that in chronic cervicitis tissues, the differences were significant(all P<0.05),moreover the positive expressions were much lower or absent in cervical cancer tissues with ⅡB-Ⅳ stage,depth of invasion>1/2 and with lymph node metastasis, the differences were significant(all P<0.05). ②The positive rates of NF-κB p50 in cervical carcinoma,CINⅠand CIN Ⅱ-Ⅲ tissues were much higher than that in chronic cervicitis tissues(all P<0.05),moreover the positive expressions were much higher in cervical cancer tissues with tumor size≥4 cm,poor differentiation,depth of invasion>1/2 and with lymph node metastasis, the differences were significant(all P<0.05). ③The expression level of BRMS1 was negatively correlated with that of NF-κB p50 in cervical carcinoma(rs= 0.426,P<0.001). Conclusions:It is closely related to cancer cell invasion and metastasis that the expression of BRMS1 decreased significantly,while the expression of NF-κB conversely increased. In addition,BRMS1 may inhibit NF-κB expression in invasion and metastasis of cervical cancer.
