ABC294640抑制卵巢癌侵袭及转移的实验研究

Inhibitory Effects of ABC294640 on Invasion and Metastasis of Ovarian Cancer

摘要: 目的:观察ABC294640对人卵巢癌细胞侵袭和迁移能力的影响,并探讨其作用机制。方法:用细胞侵袭小室法(Transwell)检测ABC294640对卵巢癌细胞侵袭和迁移能力的影响;用细胞蛋白免疫印迹(Western blotting)法分析ABC294640对卵巢癌细胞中c-Myc蛋白表达水平的调节作用。建立卵巢癌腹腔移植瘤模型,观察ABC294640对肿瘤体内转移作用的影响及其对体内c-Myc表达的影响。结果:50 μmol的ABC294640作用卵巢癌细胞后其侵袭、迁移能力受到显著抑制,细胞中c-Myc蛋白表达水平明显下降(P<0.01)。在裸鼠腹腔移植瘤模型中,与对照组比较,ABC294640用药组中腹腔转移灶数量显著减少(P<0.01)。肿瘤组织中c-Myc表达水平显著低于对照组(P<0.01)。结论:ABC294640能抑制卵巢癌细胞的侵袭和迁移能力,其抗肿瘤侵袭转移的作用机制可能与 c-Myc蛋白的表达下调有关。

Abstract: Objective:ABC294640 has shown its unique efficacy in the treatment of cancer. The purpose of this study was to investigate the inhibitory effect of ABC294640 on invasive and metastatic abilities of ovarian cancer cells and to explore its mechanism. Methods:Transwell experiments were preformed to detect the effect of ABC294640 on invasion and migration of ovarian cancer cells, and the regulatory effect of ABC294640 on the expression of c-Myc protein in ovarian cancer cells was analyzed by Western blotting. An intra-abdominal xenograft tumor model of ovarian cancer was set up to observe the effect of ABC294640 on the metastasis in vivo. Results:The ability of invasion and metastasis of ovarian cancer cells were significantly inhibited after the treatment with 50 μmol ABC294640 and the expression of c-Myc protein in ovarian cancer cells significantly decreased (P<0.01). In the intra-abdominal xenograft tumor model of ovarian cancer in nude mice, the tumor number of abdominal metastases in ABC294640 group was significantly reduced compared with the control group. The expression level of c-Myc in tumor tissue was significantly lower than that in control group (P<0.01). Conclusions:ABC294640 inhibited the invasion and migration of ovarian cancer cells, and its anti-invasion and metastasis mechanism might be related to the down-regulation of c-Myc protein expression.
