不明原因胚胎停育患者外周血及绒毛组织中芳香烃受体的表达及意义

The Expression and Significance of AhR in Peripheral Blood and Chorionic Tissues of Embryo Damage

摘要: 目的:探讨芳香烃受体(aryl hydrocarbon receptor,AhR)在胚胎停育中的作用。方法:分别选取30例不明原因胚胎停育患者作为病例组,30例正常早期妊娠人工流产妇女作为对照组。采用酶联免疫吸附试验(ELISA)方法检测外周血中AhR蛋白表达水平,实时荧光定量聚合酶链反应(PCR)方法检测绒毛组织中AhR mRNA表达水平,蛋白质印迹(Western blotting)法检测绒毛组织中AhR蛋白表达水平。结果:病例组外周血AhR蛋白表达水平[(4.99±0.82)ng/mL]高于对照组[(3.11±0.79)ng/mL],差异有统计学意义(t=2.860,P=0.046)。病例组绒毛组织中AhR mRNA表达水平(2.84±0.51)高于对照组(1.53±0.49),差异有统计学意义(t=3.208,P=0.033)。病例组绒毛组织中AhR 蛋白表达水平(0.85±0.11)高于对照组(0.61±0.09),差异有统计学意义(t=2.925,P=0.043)。结论:外周血及绒毛组织中AhR表达增高,可能与不明原因胚胎停育有关。

Abstract: Objective:To investigate the role of aryl hydrocarbon receptor (AhR) in embryo damage. Methods:30 cases with unexplained embryonic diapauses were selected as study group and 30 normal early pregnancy abortion women were selected as control group. The expression levels of AhR protein in peripheral blood were detected by enzyme-linked immunosorbent assay (ELISA). The expressions of AhR mRNA in chorionic tissue were detected by real-time fluorescence quantitative-PCR, and the expressions of AhR protein in chorionic tissue were detected by Western blotting. Results:The expression levels of AhR protein in peripheral blood of study group and control group were (4.99±0.82) ng/mL and (3.11±0.79) ng/mL, respectively. Compared with control group, the expression of AhR protein in peripheral blood of study group was significantly higher (t=2.860, P=0.046). The expression levels of AhR mRNA in chorionic tissues of study group and control group were (2.84±0.51) and (1.53±0.49). Compared with control group, the expression level of AhR mRNA in chorionic tissues of study group was significantly higher (t=3.208,P=0.033). The expression levels of AhR protein in chorionic tissues of study group and control group were (0.85±0.11) and (0.61±0.09). Compared with control group, the expression level of AhR protein in chorionic tissues of study group was significantly higher (t=2.925, P=0.043). Conclusions:The expression of AhR in peripheral blood and chorionic tissues of patients with embryo damage were significantly increased, which may be related to unexplained embryo damage.
