MS-275 on Migration and Invasion in Human Cervical Carcinoma Cell Line SiHa

Abstract: Objective:To observe the histone deacetylase inhibitor MS-275 on migration and invasion activity in human cervical carcinoma cell line SiHa,and to investigate the possible mechanism. Methods:SiHa cells were treated with different concentration of histone deacetylase inhibitor MS-275. The cell migration was estimated by wound-healing assay and the cell invasion was estimated by Transwell assay. Furthermore,Western blot was used to detect the acetyl level of histone H4 and real time PCR was used to detect mRNA of NF-κB and uPA gene. Results:After treatment with MS-275,With the increasing of concentration,the SiHa cell migration and invasion activity decreased. Meanwhile,the acetyl level of histone H4 increased,and the mRNA of NF-κB and uPA decreased in SiHa cells. Conclusions:The MS-275 could effectively inhibit the cell migration and invasion activity of SiHa cells,and the acetyl level of histone increasing to inhibite NF-κB and uPA gene expression may be one of its mechanisms.
