Application of Gonadotrophin-Releasing Hormone Analogs in Malignant Gynecological Neoplasms

Abstract: According to its mechanisms of action,gonadotrophin-releasing hormone analogs (GnRH-A) are divided into two categories:agonist and antagonist. It has been demonstrated that 80% of ovarian and endometrial cancers express GnRH and its receptors. The anti-cancer role of GnRH-A is played either by suppressing the hypothalamic-pituitary axis or through its autocrine and paracrine systems. GnRH-A had been used in the treatment of advanced or recurrent endometrial and ovarian cancers,but the effect is no better than other second-line drugs.These days,the application of GnRH-A in the preservation of fertility and ovarian function has become increasingly popular. Besides,the use of GnRH-A in molecular targeted therapy is also promising.
